en ارزیابی حجم تومورهای تجربی در موش با استفاده از سیستم تصویربرداری نوری در مراحل مختلف رشد Determination of experimental Tumor Volume of a mouse using optical imaging system in different processing stages عمومى General پژوهشي Research مقدمه: تصویربرداری مولکولی فلئورسنس در مد بازتابشی برای ارائه اطلاعات درمورد فرآیندهای پاتولوژیک یک روش قدرتمند است. رشد تومور یک فرآیند پاتولوژیک شایع است که میتوان آن را به‌وسیله کولیس اندازه‌گیری کرد. دقت اندازه‌گیری هنگام استفاده از کولیس به مهارت کاربر بستگی دارد. تحقیقات اخیر نشان میدهد که روش تصویربرداری مولکولی فلئورسنت ( Fluorescence molecular imaging (FMI) ) &lrm; 1 میتواند به عنوان روش دقیقتر جهت اندازه‌گیری آهنگ رشد تومور مورد استفاده قرار گیرد. هدف از ایجاد این پروژه اعتبار دادن به روش FMI جهت سنجش رشد تومور در مقایسه با روش‌های متداول اندازه‌گیری حجم تومور مانند اندازه‌گیری با کولیس است. روش بررسی: در این مطالعه با استفاده از مدل حیوانی تومور، همبستگی بین روش غیرتهاجمی فلئورسنت و روش‌های سنتی مورد ارزیابی قرار گرفت. دودمان سلولی WEHI-164 در 36 موش BALB/c که در سه گروه 12تایی تقسیم شده بودند به‌صورت زیر پوستی تزریق شد و پس از 10روز، اندازه تومور      به‌وسیله کولیس اندازه‌گیری گردید. به‌طور همزمان اندازه تومور با تزریق ماده فلئورسین در مرکز تومور توسط روش FMI اندازه‌گیری گردید. درنهایت همبستگی بین اندازه‌گیری حجم تومور توسط روش‌های سنتی و اندازه‌گیری توسط روش FMI توسط روش اسپیرمن و منحنی ROC ارزیابی شد. یافته‌ها: نتایج نشان میدهد که ضریب بستگی اسپیرمن بین حجم تومور و ارتفاع منحنی شدت فلئورسنت در گروه اول، دوم و سوم شدت به ترتیب 019/0 ، 5/0، 09/0 است. آنالیز منحنی ROC نشان میدهد که نقطه قطع حجم و وزن تومور بهترتیب 93 میلیمتر مکعب و 12/0 گرم می‌باشد. بحث و نتیجه‌گیری: در موقعیت ما، بین اندازه‌گیری رشد تومور در موش BALB/c توسط FMI با اندازه‌گیری توسط روش‌های مرسوم برای حجم و وزن، همبستگی معنی‌داری وجود ندارد. بنابراین دقت اندازه‌گیری روش FMI در مورد رشد تومور در حیوانات کوچک نیاز به اصلاح و بهبود دارد.  Background: Fluorescence molecular imaging (FMI) in reflectance mode is a powerful method for providing valuable information about pathologic processes. Tumor growth is a common pathologic process which can be traditionally obtained by caliper measurement. The caliper measurement has several major disadvantages. However FMI method can be used as a non-invasibly method. The goal of this project is validating FMI of tumor growth by comparing it to traditional tumor volume measurement method such as caliper measurement. <v:group coordorigin="6522,736" coordsize="67,0" id="_x0000_s1026" style="position:absolute; left:0; text-align:left; margin-left:435px; margin-top:49px; width:3.35pt; height:0; z-index:-251653632"> <v:shape coordorigin="6522,736" coordsize="67,0" filled="f" id="_x0000_s1027" path="m6522,736r67,e" strokecolor="#1121cc" strokeweight=".58pt" style="position:absolute; left:6522; top:736; width:67; height:0"> <v:path arrowok="t"> </v:path></v:shape><w:wrap anchorx="page"> </w:wrap></v:group><img alt="" src="file:///C:/Users/LENOVO/AppData/Local/Temp/msohtmlclip1/01/clip_image001.gif" style="width:6px; height:2px" >Material and Methods: Here we used an animal tumor model to evaluate the extent on correlation between noninvasively measured fluorescence and traditional methods. BALB/c mice received subcutaneous injection of WEHI-164 cells. The tumor size was measured. <a href="http://en.wikipedia.org/wiki/Fluorescein">Fluorescein  w</a>as directly injected in center of the tumor. Serial measurements of fluorescence intensities were performed with FMI. After imaging, the mice were sacrificed and tumor was removed for weighting. Results: The results showed that Spearman correlation coefficient between tumor volume and the maximum height of fluorescent intensity in the first, the second and the third group are 0.019, 0.5 and 0.09 respectively. The ROC analysis showed that the cutoff point of tumor volume and the weight are 93 mm3 and 0.12g respectively. Conclusion: The measurement of tumor growth in the BALB/c mice by imaging fluorescence correlates well with the traditional method of determining the tumor volume and with the increase in tumor weight that results from tumor growth.
